چکیده
Acetabularia acetabulum is a single-celled green alga which has historically been an important model system in cell biology. Here, we attempt to re-introduce A. as through the investigation of subcellular localization mRNAs. macroscopic with highly complex cellular morphology. The gigantic, and single nucleus, located at basal end, several centimeters away from umbrella-shaped tip. To better understand genetic basis morphology this alga, aimed characterize mRNA composition four different regions adult cells. Analyses quantitative gene expression data revealed high degree differentially distributed products. Gene transcripts involved photosynthesis were mainly accumulated apical cap structure, while end carrying nucleus was enriched for nuclear processes such DNA replication transcription. Cytoskeletal components, intracellular transport protein translation present throughout elongated cell. An observation that some others, often functionally related transcripts, localized large pools distinct regions. This suggests post-transcriptional regulation specific products may be mechanisms behind morphogenesis gigantic

Differential messenger RNA gradients in the unicellular alga Acetabularia acetabulum. Role of the cytoskeleton.
The unicellular green alga Acetabularia acetabulum has proven itself to be a superior model for studies of morphogenesis because of its large size and distinctive polar morphology. The giant cell forms an elongated tube (a stalk of up to 60 mm in length), which at its apical pole makes whorls of hairs, followed by one whorl of gametophores in the shape of a cap. At its basal pole, the cell exte...
متن کاملBeta-tubulin mutants of the unicellular green alga Chlamydomonas reinhardtii.
Two beta-tubulin mutants of the unicellular green alga Chlamydomonas reinhardtii have been isolated on the basis of altered sensitivity to the growth-inhibitory effect of colchicine. The two mutations of colR4 and colR15 have been found to be tightly linked, mapping to a previously unmarked site in linkage group XII. The drug-resistance phenotypes of both mutations segregated in genetic crosses...
متن کاملChloroplast membranes of the green alga Acetabularia mediterranea. II. Topography of the chloroplast membrane
The localization of the chlorophyll-protein complexes inside the thylakoid membrane of Acetabularia mediterranea was determined by fractionating the chloroplast membrane with EDTA and Triton X-100, by using pronase treatment, and by labeling the surface-exposed proteins with 125I. The effects of the various treatments were established by electrophoresis of the solubilized membrane fractions and...

The effects of various stresses on mitochondrial activity and structure within the unicellular green alga Chlamydomonas reinhardii were investigated using the fluorescent probe rhodamine 123. Within control cells, treatment with rhodamine 123 stained an intense fluorescent network, which was considered to be mitochondrial from the similarity in structure to models of mitochondria reconstructed ...
